Measuring Static Pressure Properly

Tech coming next week to measure me for a replacement system, LP Furnace and AC. Expect he will do a Manual J (hopefully!) and measure TESP. For the TESP, should all room duct vents be wide open or leave set to where they are? For clarification, we have 4 rooms that run electric dampers. Their default is 30% open (never fully closed). One room (2d Fl bedroom) manual damper closed as we do not heat it, though we do open for AC use. So open all up for the TESP measurement or not?
